Would users with this configuration in their preferences receive the email message?

Yes, because you did not select the "Only send to users opting to receiven ews and update emails".

So, my follow-on question is,
Can an user opt-out of email messages sent via XF ACP with the above configuration?

I think, but don't take it as gospel, that emails sent from the ACP bypass those settings.
I personally think of them as "oh crap" alert system ability.... so you can send a blast out in case of being hacked, etc.
They really should not be used to send newsletters and such.

Shortly said no.
As Tracy Perry said, it can be used in emergency situations this way, not to abuse people with newletters and stuff.
That's the reason that selection box is ticked by default.
