How do I remove signature block quotes?

I turned off the signature permission options ("Allow block tags (code, quote, etc)") but they are still showing, do I have to rebuild a cache or run a query of some kind?

Signature permission changes aren't retrospective. I think your only option is to remove all signatures (batch update in the ACP) and then users will have to redo them.
