I'm not totally sure, but it seems to me that your missing js/dark/taigachat.js

That is indeed what that error means, for some reason the script couldn't see the javascript file. This could be permissions or that the file actually isn't there or any number of things.

I thought I had checked everything too, and I've only just upgraded my live site thinking it was OK to do so.

Yes. After setting a single smilie to use the sprite sheet, it will break the image in the Taigachat dropdown box.
